Principal Investigator: Stephen T. McGarvey, PhD, MPH, Brown University, Providence, RI, USA MeSH: Obesity,Adiposity,Lipids,Blood Pressure https://www.ncbi.nlm.nih.gov/projects/gap/cgi-bin/study.cgi?study_id=phs000914 The research goal of this study is to identify genetic variation that increases susceptibility to obesity and cardiometabolic phenotypes among adult Samoans using genome-wide association (GWAS) methods. DNA from peripheral blood and phenotypic information were collected from 3,119 adult Samoans, 23 to 70 years of age. The participants reside throughout the independent nation of Samoa, which is experiencing economic development and the nutrition transition. Genotyping was performed with the Affymetrix Genome-Wide Human SNP 6.0 Array using a panel of approximately 900,000 SNPs. Anthropometric, fasting blood biomarkers and detailed dietary, physical activity, health and socio-demographic variables were collected. We are replicating the GWAS findings in an independent sample of 2,500 Samoans from earlier studies. After replication of genomic regions and informative SNPs in those regions, we will determine sequences of the important genes, and determine the specific genetic variants in the sequenced genes that are associated with adiposity and related cardiometabolic conditions. We will also identify gene by environment interactions, focusing on dietary intake patterns and nutrients.

dbGaP phs000914 Genome-wide Association Study of Adiposity in Samoans
This sample attributes data table includes body site where sample was collected, analyte type, tumor status, and sample plate and well information.
- StudyEvent: SEV1
- Eligibility Criteria
- This subject consent data table contains subject IDs, consent group information, and subject aliases.
- This subject sample mapping data table includes a mapping of study subject IDs to sample IDs. Samples are the final preps submitted for genotyping, sequencing, and/or expression data. For example, if one patient (subject ID) gave one sample, and that sample was processed differently to generate 2 sequencing runs, there would be two rows, both using the same subject ID, but having 2 unique sample IDs. The data table also includes sample aliasese and sample use.
- This subject phenotype data table includes eligibility and enrollment information (n=6 variables), relationship and twin status, anthropometric measurements (n=21 variables; height, weight, BMI, circumference, skinfold test, obesity classifications, body fat measurements, resistance, and reactance), blood pressure and heart rate (n=5 variables), diagnosis (n=14 variables; hypertension, diabetes, angina, heart attack, stroke, and heart disease), glucose and cholesterol measurements (n=21 variables; glucose, insulin, leptin, adiponectin, cholesterol, TG, VLDL, LDL, and HDL), smoking status (n=19 variables), treatment and medications for hypertension, diabetes, and heart disease (n=19 variables), and physical activity measurements (n=54 variables; work, walking, sitting, and leisure).
- This sample attributes data table includes body site where sample was collected, analyte type, tumor status, and sample plate and well information.
